Use: Media recommended for the detection of fecal coliforms in water samples by the membrane filtration technique at elevated temperatures.
Description: m FC Agar was formulated to detect fecal coliforms obtained from the intestinal tract of warm-blooded animals when using an incubation temperature of 44.5 ± 0.2°C. Nonfecal coliforms were not detected using this method. The addition of Rosolic Acid allows coliform colonies to turn blue while other microorganisms give grey to cream-colored colonies.

- Lactose ............................................. 12.5g
- Bile Salts #3 ............................. 1.5g
- Aniline Blue ......................................... 0.1g
- Agar .........................................15.0g
Final pH: 7.4 ± 0.2 at 25.0°C

Preparation: Mix 5.2 grams of the medium in a 100 ml of purified water containing 1ml of 1% rosolic acid in 0.2 N NaOH. Heat with repeated stirring and boil one minute. Cool to 45-50°C and pour plates. DO NOT AUTOCLAVE.
Quality Control Specifications:
- The powder is homogeneous, free flowing and light beige.
- With 1% rosolic acid the prepared medium is cranberry red, and slightly opalescent. Without 1% rosolic acid the medium is slightly opalescent and blue.
- Expected cultural response after 22-26 hours at 44.5°C.

Storage: Store the sealed bottle containing the dehydrated medium at 2 to 30.0°C. Once opened and recapped, place the container in a low humidity environment at the same storage temperature. Protect it from moisture and light. The dehydrated medium should be discarded if it is not free flowing or if the color has changed from the original light beige color.
