(AB) Terrific Broth Modified 500g

Terrific Broth is a highly enriched medium developed by Tartoff and Hobbs to improve yield in plasmid bearing Escherichia coli. Recombinan starins have an extended growth phase in the medium. The addition of extra Tryptone and Yeast Extract in the medium allows higher plasmid yield per volume. Glycerol is used as a carbohydrate source in this formulation. Unlike glucose, glycerol is not fermented to acetic acid. Terrific Broth is used with Glycerol in cultivating recombinant strains of Escherichia coli. (Alpha Biosciences # T20-114M-500gm)

Formula* per Liter:

• Casein Digest Peptone ......... 12.0g
• Potassium Phosphate, Monobasic... 2.2g
• Yeast Extract ........................... 24.0g
• Potassium Phosphate, Dibasic......... 9.4g..

Final pH: 7.2 ± 0.2 at 25.0°C

Preparation: Mix 47.6 grams of the medium in one Liter of purified water until evenly dispersed. Add 4mLof Glycerol to the mixture. Heat with repeated stirring and boil for one minute to dissolve completely. Distribute and autoclave at 121.0°C for 15 minutes.

Quality Control Specifications:

1. 1. The powder is homogeneous, free flowing and light beige.
2. 2. Visually the prepared medium is light to medium amber, and clear.
3. 3. Expected cultural response after 18-24 hours at 35.0°C.

Storage: Store the sealed bottle containing the dehydrated medium at 2 to 30.0°C. Once opened and recapped, place the container in a low humidity environment at the same storage temperature. Protect it from moisture and light. The dehydrated medium should be discarded if it is not free flowing or if the color has changed from the original light beige color.
