Use: Agar medium which meets APHA specifications for obtaining the microbial content of food, dairy products, water and wastewater.
Description: Trypto Glucose Extract Agar is formulated according to the specifications described by the APHA for use in performing the Aerobic Plate Count of bacteria in dairy and food products. It is also can be used for the Heterotrophic Plate Count for water and wastewater analysis.
Formula* per Liter:
- Tryptone ......................................... 5.0g
- Dextrose ............................. 1.0g
- Beef Extract ................................... 3.0g
- Agar ...................................15.0g
Final pH: 7.0 ± 0.2 at 25.0°C
* Grams per liter may be adj. or formula supplemented to obtain desired performance
Preparation: Mix 24 grams of the medium in one Liter of purified water until evenly dispersed. Heat with repeated stirring and boil for one minute to completely dissolve. Distribute and autoclave at 121.0°C for 15 minutes. Do not hold medium at 45°.0 to 50.0°C for more than 3 hours. Remelt only once.
Quality Control Specifications:
- The powder is homogeneous, free flowing and light beige.
- Visually the prepared medium is clear to slightly hazy and light to medium amber.
- Expected cultural response after 18-24 hours at 35.0°C.

Storage: Store the sealed bottle containing the dehydrated medium at 2 to 30.0°C. Once opened and recapped, place the container in a low humidity environment at the same storage temperature. Protect it from moisture and light. The dehydrated medium should be discarded if it is not free flowing or if the color has changed from the original light beige color.
