Use: Trypto Soy Agar is a general growth medium prepared with or without blood or enrichments. It is used for the isolation of different strains of fastidious microorganisms.
Description: Trypto Soy Agar (Soybean-Casein Digest Agar) is a general purpose agar medium which may be used for the cultivation of fastidious and non- fastidious aerobic and anaerobic microorganisms. A blood agar medium may be prepared by adding 5 to 10% sterile defibrinated blood. TSA with 5% sheep blood is useful for the isolation and determination of hemolytic reactions of Staphylococcus sp., Streptococcus sp. and other pathogenic microorganisms found in the upper lower respiratory tract, blood, cerebrospinal fluid, minor lesions and other clinical specimens.

Casein Digest Peptone ................ 15.0g
Agar ............................................. 15.0g
Papaic Digest of Soybean Meal.... 5.0g
Sodium Chloride ........................... 5.0g
Final pH: 7.3 ± 0.2 at 25.0°C

Preparation: Mix 40.0 grams of the medium in one Liter of purified water until evenly dispersed. Heat with repeated stirring and boil for one minute to dissolve completely. Distribute and autoclave at 121.0°C for 15 minutes. To prepare 5 to 10% blood agar by adding the appropriate volume of sterile defibrinated blood to melted sterile agar medium cooled to 45.0-50.0°C. For chocolate agar Prepare single strength agar medium and cool to 45.0-50.0°C after sterilization. Add 5 to 1O% sterile defibrinated blood and heat to 80.0°C for 10 minutes.
Quality Control Specifications:
- The powder is homogeneous, free flowing and light beige.
- Visually the prepared medium is clear to slightly hazy, yellow beige in color and free of sediment. Blood agar appears bright blood red and opaque.
- Expected cultural response after 18-24 hours at 35.0°C.

Storage: Store the sealed bottle containing the dehydrated medium at 2 to 30.0°C. Once opened and recapped, place the container in a low humidity environment at the same storage temperature. Protect it from moisture and light. The dehydrated medium should be discarded if it is not free flowing or if the color has changed from the original light beige color.
