Use: Yeast Nitrogen Base is used for classifying yeasts based on carbon assimilation.
Description: Yeast Nitrogen Base contains all essential nutrients and vitamins necessary for the cultivation of yeasts except a source of carbohydrate.

Preparation: Prepare a 10X solution by dissolving 6.7 grams of Yeast Nitrogen Base and 5 grams of Dextrose or equivalent amount of other carbohydrate in 100ml distilled or deionized water. Warm slightly to dissolve. Mix well. DO NOT BRING TO A BOIL. DO NOT AUTOCLAVE. Filter-sterilize the solution. Store at 2-8°C. Prepare the final medium by aseptically pipetting 0.5ml of the 10X solution into 4.5ml of sterile distilled water. Mix the solution thoroughly by shaking before inoculation.
Quality Control Specifications:
- The powder is homogeneous, free flowing and off-white.
- Visually the prepared medium is clear and almost colorless.
- Expected cultural response after 2-5 days at 25.0°C.

Storage: Store the sealed bottle containing the dehydrated medium at 2 to 30.0°C. Once opened and recapped, place the container in a low humidity environment at the same storage temperature. Protect it from moisture and light. The dehydrated medium should be discarded if it is not free flowing or if the color has changed from the original off-white color.
